Netrin/DCC


​Netrin-1 is a canonical axon guidance molecule best known for its ability to steer commissural axons towards the ventral midline of the spinal cord. We and others have found that Netrin-1 is also essential for many other developmental events, including morphogenesis of the semicircular canals in the inner ear and formation of a boundary between the PNS and CNS. To understand how Netrin-1 achieves such versatility, we are collaborating with X-ray crystallographers to identify ways that Netrin-1 changes its interactions with its receptor, DCC, such that signaling is sometimes permissive and sometimes instructive. We use CRISPR/cas-9 to create mice harboring structure-inspired mutations in DCC and then characterize phenotypes in a range of cellular contexts.
